ABSTRACT

Objective?To observe the clinical effect of FURS combined with ultrasound guided all seeing needle treatment of low pole renal calculi.?Methods?From January 2016 to January 2017, 63 patients with low pole renal calculi were enrolled in this study. 16 patients with multiple stones and 47 cases with single stones were treated by FURS combined with ultrasound guided all seeing needle. We retrospectively analyzed all the patients clinical data, including the operation time, bleeding volume, stone clearance rate, the incidence of postoperative complications, then assess the safety and effectiveness of the surgical approach.?Results?The operation time was 30 ~ 60 min, the average time was 45 min, postoperative complications in 3 cases, 2 cases of pain; no blood transfusion; 2 cases of percutaneous nephrolithotomy. Postoperative hospital stay wad 2 ~ 3 d, an average of 2 d, postoperative residual stone in 3 cases, 2 weeks after the treatment of external lithotripsy, 1 case of stone clearance rate of 100.0%.?Conclusion?Retrograde FURS combined with ultrasound guided visual puncture for renal calculi is safe, effective and feasible.

ABSTRACT

Objective To explore the relationship of chemokines CXCL10-135G/A and CXCL12 -801G/A gene polymorphisms with susceptibility to tuberculosis. Methods CXCL10-135G/A and CXCL12-801G/A polymorphisms of 102 tuberculosis patients(case group)and 115 healthy controls(control group)were genotyped by polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP), and the relationship between the two polymorphisms and susceptibility to tuberculosis were analyzed. Results The genotype analysis of CXCL10-135G/A and CXCL12-801G/A was in accord with the law of Hardy-Weinberg equilibrium in the case group and the control group. The differences of genotype and allele distribution frequency of CXCL10-135G/A were statistically significant between the case group and the control group(all P<0.05).The frequency of G allele distribution was higher in the case group than that in the control group, and the frequency of A allele distribution was lower than that in the control group.There were no significant differences in genotype and allele distribution frequency of CXCL12-801G/A polymorphism between the case group and the control group (all P>0.05).Conclusion Chemokine CXCL10-135G/A gene polymorphism is associated with susceptibility to pulmonary tuberculosis,and CXCL12-801G/A gene polymorphism may not be associated with tuberculosis infection.

ABSTRACT

<p><b>OBJECTIVE</b>To construct pBIFC-VN173-CXCR4 and pBIFC-VC155-NT21MP eukaryotic expression plasmids and to investigate the interaction of chemokine receptor 4 (CXCR4) and viral macrophage inflammatory protein-II(vMIP-II) N terminal 21 peptides (NT21MP) in living cells.</p><p><b>METHODS</b>DNA fragment encoding NT21MP was chemically synthesized and inserted into BiFC eukaryotic expression vector pBIFC-VC155. The full length of CXCR4 DNA fragment was amplified by RT-PCR from SKBR (3) cells and inserted into BiFC eukaryotic expression plasmid pBIFC-VN173. Two recombinant vectors were identified by restriction enzyme digestion and DNA sequencing. The recombinant vectors were cotransfected into Africa green monkey kidney fibroblast COS-7 cells by using Lipofectamine 2000. The interaction of NT21MP and CXCR4 was detected by bimolecular fluorescence complementation (BiFC) assay.</p><p><b>RESULTS</b>The restriction enzyme digestion and DNA sequences and open read frames of two vectors were consistent with experiment design. The BiFC plasmids were successfully cotransfected into the target cells and expressed. The strong BiFC signals were detected in pBIFC-VN173-CXCR4 and pBIFC-VC155-NT21MP cotransfected cells and the fluorescence signal was located in the cytoplasm.</p><p><b>CONCLUSION</b>The eukaryotic expression plasmids for BiFC assay are successfully constructed. The interaction of NT21MP and CXCR4 in living cells can be detected by using this technology.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To evaluate efficacy of closed reduction and percutaneous hollow screw fixation with prototypical retractor in treating calcaneal fracture.</p><p><b>METHODS</b>From January 2009 to June 2011, 39 patients (43 feet) with calcaneal fracture were treated by closed reduction and percutaneous hollow screw fixation with prototypical retractor. There were 33 males and 6 females, aged from 19 to 61 years with an average of 36.7 years. According to type of Sanders, type II were in 19 feet, type III were in 24 feet. Preoperative and postoperative X-ray were estimated, the data of height, width, Bölher angle, Gissane angle of calcaneous were collected, and ankle function were estimated according to AOFAS system.</p><p><b>RESULTS</b>All patients were followed up from 6 to 36 months with an average of (15.4 +/- 3.1) months. All wounds were healed well, no skin edge necrosis and infections occurred. Before operation, the height of calcaneous was average of (32.45 +/- 3.51) mm, width was (41.60 +/- 2.42) mm, Bölher angle was (8.64 +/- 13.2) degrees and Gissan angle was (136.35 +/- 15.23) degrees; while after operation, the height of calcaneous was average of (43.62 +/- 1.02) mm, width was (38.02 +/- 1.28) mm, Bölher angle was (26.87 +/- 5.32) degrees and Gissan angle was (120.78 +/- 5.34) degrees, and had significanty differences between preoperative and postoperative treatment (P<0.05). AOFAS score was improved from preoperative (35.64 +/- 11.23) to postoperative (76.18 +/- 9.87), and 29 cases got excellent results, 11 good and 3 fair. COMCLUSION: Closed reduction and percutaneous hollow screw fixation auxiliary by the retractor for the treatment is a good way, which has advantages of simple operation, satisfactory reduction fixation, reliable fixation, minimally invasive, less complications and rapid recovery.</p>

ABSTRACT

Background Our previous study demonstrated that curcumin can induce the apoptosis of retinal pigment epithelial (RPE) cells and herein inhibit the proliferation of RPE cells,and it is proved that the intravitreous injection of 0.1mg curcumin has less adverse effect to ocular tissue, inferring a good applicative prospect in clinic. Objective The goal of this experiment was to evaluate the effectiveness of curcumin on the prevention and treatment of experimental proliferative vitreoretinopathy (PVR). Methods PVR models were induced by injection of 0.1ml RPE cells (containing 2×106 cells) into vitreous cavity in 40 eyes of 20 healthy and mature New Zealand albino rabbits.0. 1ml curcumin(0. 1 mg) was then injected into lateral eye of each model rabbit immediately following the injection of RPE cells,and the equal volume of normal saline solution containing 0. 5‰ DMSO was injected into the fellow eye of each model rabbit as controls. On 1,3,7,14,21 and 28 days after injection, the changes of cornea, aqueous humor, lens, vitreous and fundus were examined and recorded by slit lamp biomicroscope, indirect ophthalmoscope,fundus color camera and B-type ultrasonograph to evaluate the inflammatory response. The incidence rate of retinal detachment was calculated and compared between curcumin group and control group. Results The inflammatory reaction in anterior chamber and misty opacity in vitreous were found from 1 day through 3 days after injection, but no obvious proliferative strap and retinal detachment in all of the experimental eyes. On the 7th day after injection, inflammatory reaction was extinct in the anterior chamber of rabbit eyes, and proliferative strap occurred in 14 eyes(75% ) in the control group but only 2 eyes (10% ) in curcumin group,showing significant difference between these two groups (P<0. 01). No retinal detachment was seen in both the two groups. On 14,21 and 28 days after injection, the incidence rate of retinal detachment was 55% ,80% ,95% respectively in control group and that of curcumin group was 10% ,15% ,15% respectively,presenting considerably differences between two groups (P<0. 01, P<0. 01 ,P<0. 01 ). Conclusion Injection of curcumin into vitreous cavity can effectively inhibit the occurrence and development of PVR in rabbit.
